I definitely enjoyed this more than the first book, not because the first book was bad--but because of how much I loved the movie first.
Who P-P-P-Plugged Roger Rabbit is an amazing blend of both worlds, taking the universe and its rules from the original novel (Toons can die, Word Balloon Speech, the comic strips, etc.) and combined it with the continuity and tone of the movie. It's still got the constantly quipping Eddie from the first, but there's a more light hearted edge that that Bob Hoskins' performance most likely influenced.
I think if you enjoyed either the first novel, or the movie, there's something here to enjoy. Eddie's comedic detective musings (and as many synonyms for 'alcohol' as possible) are always enjoyable, and draw out a laugh on occasion coupled with Roger's absurdity leave for fun time.

The plot is straight forward, and a basic 'who-done-it' that has a fun twist at the end. I didn't see it coming, but I wasn't all that surprised either; so a nice compromise. I think the Gone With The Wind tie-in, and Clark Gable's cameos were particularly enjoyable. Nothing more fun than picturing Clark Cable and Roger Rabbit arguing.
Though honestly, I think the book was worth reading just to see Eddie get the girl. But that's just me. :)
I recommend it. Good time, great characters, and as Eddie states on the first page--don't get too hung up on continuity. ;)
